What is the Handcannon?
The Handcannon is a secret obtainable pistol weapon in Resident Evil 4 Remake. It is among the best weapons in game, being able to output massive damage per shot. The Handcannon’s performance truly shines when targeting enemy weak points such as the head or legs. Despite boasting high firepower, the weapon has huge recoil so missing may prove costly, especially when up against powerful foes like the Chainsaw Man, Regenerators, Iron Maidens, and many of the game’s bosses.
Appearance-wise, the Handcannon is essentially a .50 Magnum Revolver and is one of many recurring revolver type pistols throughout the Resident Evil series.
Unlocked by clearing Professional difficulty without using bonus weapons (Primal Knife or Chicago Sweeper)
To get the Handcannon, you must beat the game on Professional difficulty without using the Primal Knife or Chicago Sweeper bonus weapons (must not be in NG+) . To access Professional mode, you will need to complete the main story for the first time on any difficulty. Afterwards, the weapon will become available for purchase from Extra Content Shop (Weapons) by exchanging Challenge Points (CP).

Handcannon Infinite Ammo
To get infinite ammo for the Handcannon, you must first purchase all tune-up (upgrade) stages for the weapon. Afterwards, the Exclusive weapon upgrade (ammo never runs out) will become available on the tune-up screen. Note that you can skip the initial requirements by purchasing Exclusive Upgrade Tickets using Spinels.

Other Gun Effects and Mechanics
Uses Ultimate Mighty Z ammo.
While yet to be confirmed, it is expected that the Handcannon will require Ultimate Mighty Z bullets to use which are quite scarce throughout the game. Because of this, it is recommended to only use the Handcannon for dealing with strong enemies and bosses as the ammo will be wasted on regular foes like the various Ganado roaming the village.
