How to Unlock Culex 3D Secret Boss Fight in Super Mario RPG Remake
Becomes accessible in post-game after obtaining the Extra Shiny Stone (key item).
To unlock the secret superboss rematch battle against Culex 3D in Super Mario RPG Remake, you must obtain the Extra Shiny Stone during the game’s post-game stages. Note that the Extra Shiny Stone is different from the Shiny Stone which only lets you battle the original 2D version of the secret boss who is much weaker than the 3D version.

How to Get Extra Shiny Stone
Rewarded for defeating Duel-Ready Johnny (Boss Rematch battle).
The Extra Shiny Stone is obtained after beating the Boss Rematch version of Johnny called Duel-Ready Johnny at the Sunken Ship. This one-on-one fight between Mario and a more powerful Johnny is only accessible during post-game as well and is not an encounter to be taken lightly.
Be sure to organize your allies to cheer for Mario during the duel as each party member can grant Mario unique buffs during the fight.

Culex 3D Boss Location
Monstro Town (Sealed Door)
After getting the Extra Shiny Stone, head to the sealed door in Monster Town. As you examine the door, the Extra Shiny Stone should start “reacting.” Afterwards, you can begin the fight against Culex 3D after entering the sealed door.
A weaker version of Culex 3D called Culex 2D can also be fought by entering the same sealed door in Monstro Town prior to beating the game. Instead of the Extra Shiny Stone won from Johnny’s Boss Rematch Battle, you will need the regular Shiny Stone to fight him.

Culex 3D Superboss Battle
A More Powerful Version of Culex 2D
Culex 3D is the hardest fight in the entire game and far surpasses the challenge posed by his 2D version in terms of HP and damage output. The number of turns you take against Culex 3D is also accounted for, though this does not have any effect on the result after defeating him.
As with the original Culex fight, Culex 3D will be accompanied by enhanced versions of the Elemental Crystals who will all bombard your party with powerful spells and debuffs.

Rewards for Beating Culex 3D
Upon defeat, Culex 3D will reward you with the Crystal Shard which serves no actual use in the game apart from being proof that you have bested the incredibly powerful version of the superboss.
As the final hurdle in the remake towards completing the Monster List and getting 100% completion, be sure to come to the fight as prepared as you can be. You will need to be at a significantly high level while using the best weapons, armor, and accessories to take down this superboss.
It is also recommended to stock up on as many healing items as you can to improve the party’s staying power throughout the fight.
Attain “Mario Sage” status after clearing all Boss Rematch battles.
Apart from the rewards received for clearing each Boss Rematch (including the final rematch against Culex 3D), you do not actually get any major prize once all of them have been completed. Frog Sage will simply start referring to you as “Mario Sage” onwards when talking to him as recognition of your strength in battle.
