Shisenin Kosen is an optional Dojo Boss in Nioh. You will encounter the boss in the Dojo Mission A Clash of Sickles, which is a mission that you’ll unlock if you have a 500,000 proficiency with the Kusarigama. You must also complete the Main Mission Immortal Flame to unlock this Dojo Mission.

Shisenin Kosen Boss Guide
This fight is fairly easy if you know how to use your kusarigama well. The best way to defeat the boss easily is to deplete its Ki, so hit it with strong kusarigama attacks, then do grapples and ground finishers for bigger damage. Block his attacks then strike him after his barrage.
Defeating Shisenin unlocks the mystic art for the dual katanas and a Kusarigama Master Trophy.
